Dexter, tytułowy główny bohater serialu, w ciągu dnia pracujący w policji gdzie jest odpowiedzialny za analizę krwi. Jednak po godzinach ukazuje swoje drugie oblicze – tropi tych, którym udało się uniknąć kary i własnoręcznie wymierza im sprawiedliwość. W dzieciństwie adoptowany przez policjanta Harry’ego, który nauczył go zaspokajać swoje chore pragnienia. Od czasu śmierci swojego przybranego ojca, nikt nie poznał jego mrocznej tajemnicy, nawet jego przybrana siostra Debra. Pewnego dnia w jego życie wkrada się morderca z ciężarówki-chłodni (nazwany tak przez policję), który wzbudza w nim podziw – okalecza on swe ofiary w wyjątkowy sposób. Okazuje się, że zabójca wie wiele o życiu Dextera i o jego sekrecie.
